Agaricus brasiliensis polysaccharides stimulate human monocytes to capture Candida albicans, express toll-like receptors 2 and 4, and produce pro-inflammatory cytokines J. Venom. Anim. Toxins incl. Trop. Dis.
Martins,Priscila Raquel; Soares,Ângela Maria Victoriano de Campos; Domeneghini,Andrea Vanessa da Silva Pinto; Golim,Márjorie Assis; Kaneno,Ramon.
Abstract Background Agaricus brasiliensis is a medicinal mushroom with immunomodulatory and antitumor activities attributed to the β-glucans presented in the polysaccharide fraction of its fruiting body. Since β-glucans enhance cellular immunoresponsiveness, in this study we aimed to evaluate the effect of an acid-treated polysaccharide-rich fraction (ATF) of A. brasiliensis on the ability of human monocytes to adhere/phagocyte C. albicans yeast cells, their expression of pattern recognition receptors and their ability to produce cytokines. Methods Adhesion/phagocytosis of FITC-labeled C. albicans was evaluated by flow cytometry. Cells were incubated with specific fluorochrome-labeled antibodies for TLR2 and 4, βGR and MR and also evaluated by flow...

In vitro generation of human monocyte-derived dendritic cells methodological aspects in a comprehensive review Anais da ABC (AABC)
CECHIM,GIOVANA; CHIES,JOSÉ A.B..
Abstract: Dendritic cells (DCs) initiate and shape both innate and adaptive immune responses. They are specialized in antigen presentation to naive T cells, thereby orchestrating the T cell immune responses. Human peripheral blood and tissues contain several subsets of phenotypically and functionally distinct DCs, which promote interactions between the external environment and lymphoid organs. Because of the difficulty in purifying these cells, in vitro studies only became more frequent when Frederica Sallusto and Antonio Lanzavecchia developed a method to generate DCs from blood monocytes in vitro. Nowadays a wide range of biotechnological innovations has allowed the study of DCs and their precursors in the most diverse situations faced by the immune...

Leptospira interrogans activation of peripheral blood monocyte glycolipoprotein demonstrated in whole blood by the release of IL-6 BJMBR
Dorigatti,F.; Brunialti,M.K.C.; Romero,E.C.; Kallas,E.G.; Salomão,R..
Glycolipoprotein (GLP) from pathogenic serovars of Leptospira has been implicated in the pathogenesis of leptospirosis by its presence in tissues of experimental animals with leptospirosis, the inhibition of the Na,K-ATPase pump activity, and induced production of cytokines. The aims of the present study were to investigate the induction of IL-6 by GLP in peripheral blood mononuclear cells (PBMC) and to demonstrate monocyte stimulation at the cellular level in whole blood from healthy volunteers. PBMC were stimulated with increasing concentrations (5 to 2500 ng/ml) of GLP extracted from the pathogenic L. interrogans serovar Copenhageni, lipopolysaccharide (positive control) or medium (negative control), and supernatants were collected after 6, 20/24, and...

Lipopolysaccharide-induced expression of cell surface receptors and cell activation of neutrophils and monocytes in whole human blood BJMBR
Gomes,N.E.; Brunialti,M.K.C.; Mendes,M.E.; Freudenberg,M.; Galanos,C.; Salomão,R..
Lipopolysaccharide (LPS) activates neutrophils and monocytes, inducing a wide array of biological activities. LPS rough (R) and smooth (S) forms signal through Toll-like receptor 4 (TLR4), but differ in their requirement for CD14. Since the R-form LPS can interact with TLR4 independent of CD14 and the differential expression of CD14 on neutrophils and monocytes, we used the S-form LPS from Salmonella abortus equi and the R-form LPS from Salmonella minnesota mutants to evaluate LPS-induced activation of human neutrophils and monocytes in whole blood from healthy volunteers. Expression of cell surface receptors and reactive oxygen species (ROS) and nitric oxide (NO) generation were measured by flow cytometry in whole blood monocytes and neutrophils. The...

Superoxide release and cellular gluthatione peroxidase activity in leukocytes from children with persistent asthma BJMBR
Marçal,L.E.; Rehder,J.; Newburger,P.E.; Condino-Neto,A..
Asthma is an inflammatory condition characterized by the involvement of several mediators, including reactive oxygen species. The aim of the present study was to investigate the superoxide release and cellular glutathione peroxidase (cGPx) activity in peripheral blood granulocytes and monocytes from children and adolescents with atopic asthma. Forty-four patients were selected and classified as having intermittent or persistent asthma (mild, moderate or severe). The spontaneous or phorbol myristate acetate (PMA, 30 nM)-induced superoxide release by granulocytes and monocytes was determined at 0, 5, 15, and 25 min. cGPx activity was assayed spectrophotometrically. The spontaneous superoxide release by granulocytes from patients with mild (N = 15), moderate...
